For travelers the best option can be taking a flight from CCU or Kolkata Airport to Gopinath Bordoloi Airport, Assam or to Umroi Airport in Meghalaya. Several flights are available from Kolkata airport to Assam & Meghalaya every week.

From my experience, it is better to book tickets for Guwahati airport, Assam, as because flights can suddenly get cancelled due to poor visibility and bad weather in Meghalaya. Also larger jets are not permitted to Meghalaya due to hilly terrains and sudden weather fluctuations, thus you get to travel only in smaller turboprop planes such as the ATR 72 which takes time and is noisy to the ears.

Travelers can easily journey from Guwahati Airport to Meghalaya in less than four hours, as there are a plethora of cars available for rental to other places, Shillong being one of the most common places.

Taxi fares range from INR 1600 to INR 5000 from entry level to high end cars. The distance is roughly 130 KM. You can stop for food along the various highway shops you will find along the way.
